Novel treatments for chronic disease

ARYx currently has four proprietary drug candidates in the clinic and additional leads undergoing optimization. While our technologies can be applied to oral drugs, topical agents and injectables for a wide variety of disease indications, our initial candidates are oral drugs aimed at large, chronic markets. ATI-7505 - Gastrointestinal Disorders

ATI-7505 is a proprietary serotonin type 4 (5HT4) agonist in Phase 2 development for the treatment of multiple gastrointestinal disorders. Potential indications include chronic constipation, functional dyspepsia, gastroesophageal reflux disease (GERD), gastroparesis (delayed emptying of the stomach), and inflammatory bowel disease with constipation. Each of these indications addresses significant patient populations. The efficacy of 5HT4 agonists in gastrointestinal disorders is well established, although serious cardiac liabilities have significantly limited their use. ATI-7505 is designed to provide prokinetic activity in the gastrointestinal tract without cardiac safety problems at anticipated therapeutic doeses.

Budiodarone (ATI-2042) - Atrial Fibrillation

Budiodarone is a proprietary compound in Phase 2 development for the treatment of atrial fibrillation (AFIB). ARYx has engineered budiodarone with the goal of developing a therapy equally effective but safer than amiodarone, considered by physicians to be the most effective drug for treating patients with atrial fibrillation. Although amiodarone is not approved for treatment of atrial fibrillation, it remains the market leader. Treatment of AFIB with amiodarone has been severely limited by the toxic side effects resulting from accumulation of the drug. Budiodarone is designed to preserve the effective pharmacology of amiodarone while mitigating the safety issues.

Tecarfarin (ATI-5923) - Anticoagulation

Tecarfarin, a novel compound for oral anti-coagulation therapy, is currently in Phase 2/3 clinical trials. ARYx has designed tecarfarin to preserve the efficacy and complex pharmacology of Coumadin® (warfarin), the current market leader, while improving the safety profile. Like warfarin, tecarfarin depletes certain clotting proteins in the blood, thereby reducing the occurrence of deep vein thrombosis (DVT) and the risk of stroke associated with atrial fibrillation (AFIB). Tecarfarin is metabolized by non-oxidative enzymes to a single inactive entity and thus far appears to have less variable metabolism and reduced instances of under/over anticoagulation, making the response to tecarfarin safer and more predictable.

ATI-9242 - Psychiatric Disorders

ATI-9242 is designed to be a novel, next-generation atypical antipsychotic for the treatment of schizophrenia and other psychiatric disorders. We believe this product will improve upon the efficacy of the best atypicals while avoiding metabolic drug-drug interactions and minimizing certain other metabolic problems associated with atypicals, including weight gain and type 2 diabetes. The receptor profile of ATI-9242 is designed to treat negative symptoms of schizophrenia - reduction in social interaction, disassociation from people or settings, monotone speech, loss of feelings of pleasure - and enhance cognitive functions. We entered the clinic with ATI-9242 in April 2008.
